    In his efforts to elucidate the physical and chemical characteristics of atmospheric aerosols related to their adverse health effect, associate professor Tomoaki Okuda of Keio University’s Department of Applied Chemistry Faculty of Science and Technology is conducting research on three areas: aerosol collection technology, aerosol surface area, and aerosol charging state.

    “As for my motivation, I wanted to perform cellular exposure biochemical experiments using aerosol particles actually present in the air. The conventional approach is to use a filter to collect particles. But this ultimately leads to the filter becoming clogged and being unable to collect large amount of particles. Also, when performing cellular exposure experiments, material from the filter itself would have a negative effect. This was a problem, so to solve this we are employing a method of collecting particles using a cyclone. This enables a large amount of particles to be collected, and because a filter is not used, the effects of the filter are also not present, so we think this is a suitable method for cellular exposure biochemical experiments.”

    Currently there is concern regarding the health effects of PM2.5 fine particulate matter less than 2.5 microns in diameter. Associate Professor Okuda is developing a high-volume collection device to simultaneously collect PM2.5 particles as well as coarse particles including yellow sand particles. This device uses a virtual impactor, and by drawing in large volumes of air at high speed into the device, it becomes possible to separate fine particulate matter from coarse particulate matter. Subsequent use of a cyclones enables collection of each of these types of particles.

    “As for why there is a relationship between particle surface area and toxicity, it is the reaction that occurs at the surface of the particle in the living organism when it is taken into the body. For example, there is a reaction in the body that produces free radicals. There are several reports that the amount of radical production, which occurs on the particle surface, is proportional to the particle surface area. In addition, as for pollutants in the environment being introduced into the body, harmful substances that coexist in the environment stick to the surface of particles, so larger surface area makes it easier for pollutants to enter the body. Based on the two points of view, which are chemical reaction and physical adsorption that are enhanced on the particle surface, I think that particle surface area and toxicity are related.”

    In an experiment in which carbon nanotubes known as fine particulate matter were administered to mice, it was found that as the carbon nanotube surface area increased, inflammatory activity in the airway cells in mice also increased. By using a technique known as diffusion charging, Associate Professor Okuda has begun measuring the surface area of ambient aerosol particles in real time. In this technique, as aerosol particles are passed through a chamber where a large number of ions have been produced, ions become attached to the particles in proportion to particle surface area, and then the electric charge of ions is measured downstream. Currently, it is now being understood that the surface area of ambient aerosols is significantly controlled by the behavior of specific chemical components that constitute aerosols.

    I think that the electrostatic charging state of aerosols is also related to harmfulness. There have been examples of reports and experiment results on this issue in the past. Experiments have been conducted on the deposition of particles using a model of the human airway. The previous study has observed differences in particle deposition after passing charged and uncharged particles through the human airway model. According to the results, charged particles attached to the airway surface six times more than uncharged particles. If that is the case, six times as many particles would become attached to the body, so from the body’s perspective, that is six times as much chemical substances, meaning it could likely be having a significant effect on its harmfulness and a person’s health. I think the charging state of aerosol particles could likely have a significant effect on a person’s health. However, there are almost no examples of reports focusing on the actual charging state of ambient aerosols.
